Welocity Genetics carefully processes all the saliva samples in its NABL accredited cutting-edge technology DNA lab that employs the Next Generation Sequencing genetic machines imported from USA. Every single material used, may it be the genotyping chip or the reagents, are of the highest quality, to ensure authentic and accurate genetic yield to deliver gold-standard reports.
Samples arrive at our lab regularly, in batches. The first thing technicians do with each sample is to scan the barcode on it, because, for privacy reasons, that's the only identifying information the laboratory has.
As soon as a sample is scanned, it undergoes its first test - a visual inspection to make sure it contains fluid up to the "fill line" on the tube. Occasionally a sample does not pass this step, either because the customer did not fully squeeze the preservation ampoule and empty all of the content into the collection funnel or the sample got leaked in transit. In such rare cases, if a sample tube doesn't contain enough liquid, we immediately contact the customer and request resampling so that he or she can spit again.
Once a sample passes the visual inspection, it is time for the DNA extraction. This is the process our NABL accredited laboratory uses to isolate and purify the DNA from your saliva. Once the DNA is extracted from the saliva sample, the concentration is measured to make sure there is enough for the actual analysis process on the genotyping chip.
In case in the first attempt of DNA extraction does not yield satisfactory results, the sample will be re-run for a second attempt with part of the saliva that was set aside before the process was startedwith the hope that the second sample will be DNA-rich and ready to go. Even if the rerun (second attempt) fails, we contact the customer and request resampling so he or she can spit again.
There are multiple reasons for the low DNA yield. Most common cause of low DNA yield is thatsamples are contaminated with residue of foods and other contaminants that have mixed with the saliva. At times, customers do not fully release the preservative ampoule into the tube. Some people just naturally have low DNA in their saliva, which is a very rare occurrence.Any which ways, it is important to handle the sample very carefully and not contaminate it while collecting. For this, you must follow the instructions as printed on the saliva kit and mandatorily watch the sample collection video.
Samples that yield sufficient quantities of DNA are submitted for genotyping on our custom chip. This process is performed in large batches of saliva samples. We closely monitor this process to make sure it is going as expected. If a particular sample fails to give information for subset of the markers we test for, we go back to the saliva sample that was set aside in the extraction step and try again. If a second attempt at genotyping fails, we request a sample replacement to the customer so he or she can spit again.
Once a sample has been successfully analyzed, the laboratory sends the resulting genomic data along with the barcode that came with the sample to our bioinformatics team. This allows us to return the information to the proper account and begin interpreting your genetic data.
Once your genomic data is with our bioinformatics team it goes through our proprietary algorithm which is developed by our credible and highly specialized team of bioinformaticians, geneticists, scientists, health professionals and researchers that have spent years studying, understanding and mapping out the relationship between genotypes and phenotypes across many thousands of scientific publications. Once your report is fully generated, it is sent to you as a PDF by our support team by email or WhatsApp.
